268. In the present case, what had been presented to the Chair of the Board, Achmat, was a hearsay ‘rumour’ and four speculations involving innuendo called “questions”. Detectible fusion hybrids in both primary and metastatic sites supported the possibility that fused neoplastic cells readily disseminate from primary to distant sites. To explore this, we collected blood from GFP+ mice with established isogenic RFP+ B16F10 tumors (Fig. 4A). Peripheral blood was subjected to flow cytometry for quantification of circulating tumor cells (CTCs) as defined by their RFP expression. RFP+/GFP+ fusion hybrids (or CHC) were easily detectible within the total RFP+ population, representing 90.1% of the tumor cells in circulation, markedly outnumbering unfused RFP+/GFP−ve CTCs (Fig. 4A). Imaging of individual CHCs confirmed their fusion identity and morphologically distinguished them from MФs that had phagocytosed or adhered to cancer cells (Fig. 4A).

Bodywork using actual tools is by far our least favorite modification to perform. However, over the years we have done several body mods that involved welding on thin sheetmetal. Getting perfect show-quality results will require a different process than what we propose, because adding too much heat to the panel will cause it to warp. We’re not “show-quality” welders, but assuming that you are just trying to get the two metal bits to stick together without warping, the first thing to do is to check the wire diameter in your welder. If you are running 0.035- or 0.030-inch-diameter MIG wire, you’ll likely want to swap it out for some 0.025- or 0.023-inch-diameter welding wire. The thinner wire will give you the ability to put less heat into the thin sheetmetal, avoiding a blow through. You’ll need to practice and fine-tune the wire speed and voltage settings prior to actually welding on your door project. If one of the panels being welded is thicker than the other, start from the thick part and focus your heat there, then drag the weld puddle into the thinner panel and back to the thick panel. For more delicate body metal projects, you may need to use a series of small tack welds. Skip around along the seam for each tack weld to avoid putting too much heat in one area. You can work your way back and forth until the panel is fully welded. Once you are done you can grind the tack weld bead smooth and use body filler as needed to smooth the area.

B16F10 (fl-dsRed-fl-eGFP) primary tumors and lungs from LysM-Cre mice were fixed in 4% paraformaldehyde for 2 hours at 20°C, washed, and cryopreserved in 30% sucrose for 16 hours at 4°C and then embedded in OCT. Primary tumors were stained as described in the paragraph above. Lung sections were cut to 8-μm thickness, baked for 30 min at 37°C, then subjected to antigen retrieval under standard conditions (R&D Systems, CTS016), blocked with DAKO Protein Block Serum-Free (Agilent, X090930-2), and incubated for 16 hours at 4°C with primary antibodies [anti-MITF (1:500; Abcam, ab12039), anti-dsRed (1:250; Clontech, 632496), and anti-GFP (1:1000; Abcam, ab13970)] in background-reducing antibody diluent (Agilent, S302281-2). Fluorescent-tagged secondary antibodies were applied, and then sections were mounted in a ProLong Gold antifade reagent (Molecular Probes, P36934). Antibody specificity was determined by immunostaining healthy lungs of nontumor-bearing mice and performing secondary antibody only controls.

It is my view that it would have been most inappropriate of Achmat, Chairperson of Equal Education, who had handed investigation of this matter over to the Human Resources Subcommittee to have been tendering or evidence or information or guidance to the very committee entrusted with such an issue. It matters little whether or no this information could ever have been useful. It matters little whether or no this information would have assisted any person who might be giving consideration to a complaint regarding sexting. Achmat might have been intruding upon the merits of a process in which as Chair of the Board he should have played no part, in which he purported to play no part and where his position as Chair of Equal Education should have truly resulted in his recusal from any discussions leading to any findings of this hearing. This was, after all, a preliminary investigation to determine whether or not disciplinary proceedings should follow in accordance with the Code.
